The OIE/NIAH-DLD Regional Workshop for Laboratory Staff Members of HPAI Diagnostic Laboratories (Bangkok, Thailand, 26-30 March 2007)
The OIE/NIAH-DLD Regional Workshop on advanced technologies of sequencers and real-time PCR for Laboratory Staff Members of HPAI Diagnostic Laboratories was held in Bangkok, Thailand, on 26-30 March 2007.
The Workshop was attended by nine participants from Cambodia, Indonesia, Laos, Malaysia, Myanmar, Thailand and Vietnam, and three observers from Thailand. The participants were trained for sequencing and sequence analysis, and real-time PCR, at National Institute of Animal Health (NIAH), Bangkok.
Dr Monaya Ekgatat, Deputy Director NIAH, delivered an opening speech to the participants. Dr Shiro Yoshimura, OIE Senior Deputy Regional Representative for Asia and the Pacific, and Dr Chaweewan Leowijuk, Deputy Director General, DLD, also delivered welcome speeches as a co-organizer.
An introductory lecture was given by Dr Wasun Chantratita, Chief of Virology and Microbiology Unit, Department of Pathology, Faculty of Medicine, Ramathibodi Hospital, Mahidol University, Bangkok, to provide participants with an overview of sequencing and sequence analysis as well as PCR (real-time PCR and reverse transcription PCR) for HPAI diagnosis.
On the second day, lectures were delivered to give an overview of RNA extraction, cDNA synthesis, and RT/PCR for differential diagnosis. A hands-on exercise included RNA extraction from unknown samples, cDNA synthesis, RT/PCR for differential diagnosis, gel electrophoresis, and purification of PCR products.
On the third day, lectures were delivered to give an overview of Avian Influenza Virus (AIV) genetic sequencing and RT/PCR for subtyping of AIV. A hands-on exercise comprised purification of PCR products in continuation from the previous exercise, RT/PCR for subtyping, removing excess dye terminator from sequencing reactions, and sequencing reaction.
On the fourth day, lectures were delivered to give an overview of sequencing analysis of AIV. Hands-on exercises were performed on sequence analysis using personal computers.
On the fifth day, Further lectures and hands-on exercises were provided in continuation to the exercises in the previous day and evaluation session was held in the afternoon.
